Well the upgrade is if you already have Xenforo installed on your server in a previous versions and full release is if you haven't Xenforo installed yet.

can i upgrade from 2.0.1.0 to this build directly?

Yes, you may do that but be aware that its beta version and not stable. Some bugs could be there!
